The study aimed to establish the most suitable osmolarity range of the hypoosmotic solution for testing the membrane integrity in raw semen of rams. Sodium citrate and fructose based solutions (S) with the following osmolarities (mOsm/L) were used: 50 (S1), 75 (S2), 100 (S3), 125 (S4), 150 (S5), 175 (S6), 200 (S7), 250 (S8), 290 (S9) and 300 mOsm/L (S10). Nine mature Santa Ines rams (21.5+0.5 months; 72.9+1.6 kg) had semen collected one month apart. After the hypoosmotic swelling test (HOST) the ejaculates were evaluated using a phase-contrast microscope. A total of 200 spermatozoa were counted and sperm cells were classified as spermatozoa with coiled tails (CD, %), strongly coiled tail (CFD, %) or non-coiled tails. According to CD, S1, S2, S3, S4, S5, S6 and S7 were superior to S8, S9 and S10 (P<0,05). According to CED, S2, S3, S4, S5, S6 and S7 were superior to S8, S9 and S10 (P<0,05). Results suggest that the 75 up to 200 mOsm/L solution would be bestfor using in HOST when raw semen of rams is analyzed. |

Some Sciaridae species are considered as cosmopolitan pests in greenhouses and mushrooms farms. The potential harm of these insects involves plant tissue damage and transmission of pathogens to the seedlings. Recently, large sciarid masses were found infesting wood log stocking of processing industries and turned to be a severe nuisance factor and a public health issue. The Sciarid specimens from infested areas were identified belonging to the genus Bradysia Winnertz, 1867. These species certainly do not correspond to pests, but to species that are possibly associated to microenvironments generated by the fungi on the wood bark. |
