Sperm motility was examined periodically for 8 hours after thawing of washed (w) and unwashed (uw) goat semen extended with Skim milk-Glycerol (SMG), Lactose-Egg yolk-Glycerol (LEG), and Tris-Citric acid-Egg yolk-Glycerol (TEG) to determine the effect of the removal of seminal plasma. Semen from 4 bucks was collected with the artificial vagina, pooled and divided in 6 aliquots. Three (3) aliquots were washed with Ringer's solution in a centrifuge at 950 XG twice for 15 minutes. All 6 aliquots (3 w and 3 uw) were extended in SMG, LEG, and TEG to 400 X 10 6 motile sperm per ml, cooled and held at 5ÀC for 2 hours. The semen was filled into 0.25 ml French straws and frozen in liquid nitrogen vapor. The semen was thawed in a water bath at 37ÀC and kept at 37ÀC for motility evaluation at 0, 2, 4, 6 and 8 hours. The experiment was repeated 7 times. The average sperm motility immediately after thawing was 61.4 and 42.1 for SMG (w) and SMG (uw), 59.3 and 20.7 for LEG (w) and LEG (uw), and 53.6 and 47.1 for TEG (w) and TEG (uw), respectively. There was no difference between post-thawed sperm motility during the 8 hours evaluation period in samples containing 'w' or 'uw' sperm extended in SMG. However, a higher number (P less than 0.005) of motile sperm were observed in samples containing 'w' sperm than the samples containing 'uw' sperm extended in LEG of TEG. It seems necessary to remove seminal plasma from goat semen when extended in a medium containing egg yolk |
