We assessed the effect of stocking density on physiological parameters (blood lactate, glucose, cortisol, hematocrit), water quality (temperature, dissolved oxygen, pH, unionized ammonia, carbon dioxide), and survival during the transportation of fingerling (24.5 ± 4.7 g) and juvenile (615.8 ± 122.2 g) pirarucu (Arapaima gigas) for six hours in plastic bags. The tested densities were 65, 80, 95, 110 and 125 g L-1 for fingerlings, and 50, 80, 110, 140 and 170 g L-1 for juveniles (three replicates each). Parameters were measured prior to and immediately after transportation, and at 24 and 96 hours recovery after transportation. No mortality was observed, except for fingerlings (< 3%) at densities of 110 and 125 g L-1 during recovery. All the water quality parameters were significantly altered after the transportation of fingerlings and juveniles. Water temperature, dissolved oxygen, carbon dioxide and unionized ammonia increased, but pH decreased. Only carbon dioxide and unionized ammonia differed among densities. Cortisol levels did not increase over time, except for the juveniles at 170 g L-1, which still had high cortisol after 96 hours. Glucose significantly increased after transportation for all the treatments and returned to the initial values during the recovery period. Conversely, the lactate values were still high after 96 hours. Hematocrit was assessed only for juveniles and was significantly lower after transportation. We conclude that fingerling and juvenile pirarucu can be safely transported at densities up to 95 g L-1 and 140 g L-1, respectively. Resumo: As nanopartículas de prata (AgNP) liberam continuamente um baixo nível de íons de prata para fornecerem proteção contra bactérias e têm sido empregadas em revestimentos antimicrobianos, tecidos, dispositivos biomédicos e outros materiais. No setor agrícola têm sido usadas na desinfecção de sementes e bulbos, assim como no controle de crescimento de fungos como Podosphaera pannosa em abóboras e Pythium ultimum e Colletotrichum gloeosporioides em outros vegetais. A produção de AgNP e o aumento do consumo de produtos que as contém pode levar à contaminação de compartimentos ambientais. Os nematoides de vida livre fazem parte da cadeia alimentar destes compartimentos. Este estudo teve como objetivo a avaliação da toxicidade aguda da AgNP sobre o nematoide Panagrolaimus sp e a utilização deste dado para estimar um nível de proteção para invertebrados do sistema aquático e sua zona de transição com o solo. Os organismos foram expostos à nanopartículas de prata estabilizada em álcool polivinílico (AgNP- PVA) em diferentes concentrações (0,0; 0,01; 0,1; 1; 10 and 100 mg/L). Foi registrada a imobilidade e determinada a concentração efetiva média por um período de 96 horas (CE50-96h) equivalente a 8,45 mg/L. O parâmetro de toxicidade obtido foi útil para estimar uma concentração de baixo risco (2,5 x 10-6 mg/L) para invertebrados. Os resultados auxiliam no estabelecimento de concentrações máximas protetivas das comunidades, assim como no controle de lançamento de efluentes que possam conter o nanomaterial. -- Abstrcat: Silver nanoparticles (AgNP) continuously release a low level of silver ions to provide protection against bacteria and have been used in antimicrobial coatings, clothing fabrics, biomedical devices and other materials. In the agricultural sector it has been used to disinfect seeds and bulbs, as well as to control the growth of fungi such as Podosphaera pannosa in pumpkins, and Pythium ultimum and Colletotrichum gloeosporioides in other vegetables. The production of AgNP and the increased consumption of products that contain them can lead to contamination of environmental compartments. Free-living nematodes belong to the food chain in such compartments. This study aimed to evaluate the acute toxicity of AgNP particles stabilized in polyvinyl alcohol (AgNP-PVA) on the nematode Panagrolaimus sp. and the use this data to estimate a level of protection for inhabits the aquatic system and its transition zone with the soil. The organisms were exposed to AgNP particles stabilized in polyvinyl alcohol (AgNP-PVA) in different concentrations (0.0; 0.01; 0.1; 1; 10 and 100 mg/L). Immobility was recorded and the average effective concentration was determined for a period of 96 hours (EC50-96h) equivalent to 8.45 mg/L. The toxicity parameter obtained was useful to estimate a low risk concentration (2.5 x 10-6 mg/L) for invertebrates. The results help to establish maximum protective concentrations for the communities, as well as to control the release of effluents that may contain the nanomaterial.
