Endophytic diazotrophic plant growth-promoting bacteria Herbaspirillum rubrisubalbicans (HCC103), Herbaspirillum seropedicae (HRC54), Paraburkholderia tropica (Ppe8 T ), Gluconacetobacter diazotrophicus (Pal5 T ), and Nitrospirillum amazonense (CBAmC) have been used as inoculants for sugarcane. The genome sequences of these strains were used to design a set of specific primers for the real-time PCR (qPCR) assay. Primer specificity was confirmed by conventional PCR using the genomic DNAs of 25 related bacterial species and the five target strains. The qPCR assays were conducted using root and shoot samples from two sugarcane varieties (RB867515 and RB92579). These samples were collected both with and without inoculation, using the target strains specified in this study. The sugarcane plants were grown in a greenhouse, utilizing a substrate composed of sterile sand and vermiculite in a 2:1 ratio, for a duration of 55 days. The primers designed for this study successfully amplified target DNA fragments from each of the bacterial species, enabling their differentiation at the species level. The total bacterial population present in the sugarcane quantified using qPCR was on average 105.2 cells g−1 of fresh tissue. Across both evaluated varieties, it was observed that the population of inoculated bacteria tended to decrease over time and became more concentrated in the sugarcane roots compared to the aerial parts. O objetivo deste trabalho foi utilizar geotecnologias, sensoriamento remoto e sistema de informação geográfica (SIG) para mapear e avaliar as relações espaço-temporais entre o uso e ocupação das terras com café e os solos da região de São Sebastião do Paraíso, no sul de Minas Gerais, realizada entre os anos de 2000 e 2009. Para o processamento das imagens, mapeamento do uso da terra e estudo da evolução, foi utilizado o sistema para processamento de informações georreferenciadas ? SPRING. A região estudada abrange uma área de 520 km2. O estudo espaço temporal da região cafeeira de São Sebastião do Paraíso mostrou que o parque cafeeiro da região cresceu 4 % entre os anos estudados e mostrou que 39% das áreas plantadas surgiram depois do ano 2000, o que reflete um parque cafeeiro renovado. A cafeicultura na região encontra-se predominantemente no ambiente geomórfico-geopedológico oeste, onde os solos, principalmente os Latossolos Vermelhos férricos, são mais aptos ao cultivo. Surgiram novas áreas cafeeiras na porção leste da região, caracterizadas por pequenas lavouras, indicando também uma possível expansão da agricultura familiar na região O presente trabalho mostrou que as geotecnologias podem ser utilizadas para avaliar a evolução de áreas cafeeiras na região de São Sebastião do Paraíso e gerar informações para o agronegócio café. |
