Recombinant proteins are incresingly being used as therapeutic agents. Clinical applications often require large amounts of highly purified molecules, often for chronic treatments. Produciton in the milk of transgenic dairy animals is currently being tested as an alternative to plasma fractionation for the manufacture of a number of blood factors, recombinant antibodies and growth factors. The following review examines the potential of transgenic goats for the production of recombinant therapeutic proteins. To date, more than 60 proteins have been expressed in the milk of transgenic mice, rabbits, goats, and dairy cows. A factor in the generation of transgenic goats for the production of therapeutic protein in milkis the method employed to introduce the exogenous DNA in the germline. To date, the method most frequently employed to produce trangenic goats has been pronuclear microinjection. While successful and widely used, the microinjection approach has had limited efficiency.. For most proteins, expression in th range of 3 - 5 grams/ liter do not seeem to adversely affect lactation volumes. It is possible to achieve high-level expression of recombinant therapeutic proteins in the milk of transgenic animals. This technology could also be useful for the improvement of carine dairy herds, especially increasing the yield of cheese. |
