An improved new method was developed for the separation and purification of the flavonoid constituents of soyabean (Glycine max L. Merrill) leaves. The crude extract (40% EtOH) was subjected to Amberlite XAD-2 column chromatography. Sephadex LH-20, TLC, and a combination of HPLC gradient and isocratic elution made possible the separation and identification of seven flavonoid glycosides in the insect-resistant phenotype PI 227687. Quercetin and kaempferol glucosyl(1->6) galactosides are reported for first time in soyabean phenotypes and the isorhamnetin analogue also present is a new flavonoid glycoside. Quercetin, kaempferol and isorhamnetin 3- rutinosides and the isoflavone genistin were also identified from PI 227687. Crude extracts were directly testedagainst insects as well as three fractions (A,B, and C) obtained by using paper chromatography in chloroform:acetic acid:water as solvent. Additional experiments were carried out to test the effect of pure rutin and all bioassays were developed with Heliothis virescens (F.) and Trichoplusia ni (Hubner) (Lepidoptera:Noctuidae), from egg hatching to pupation. The survival of H. virescens (55%) and T.ni (65%) were affected by PI 227687 crude extracts. The growth assessment of surviving T.ni fed on the fraction-diet was similar to those insects fed on control diet probably because they fed for a longer time. The weight of larvae at day 10 and feeding and development time of both H.virescens and T.ni were adversely affected by PI 227687 fractions. H.virescens ...
