Even though much improvement has been made in plant transformation methods, the screening of transgenic plants is often a laborious work. Most approaches for detecting the transgene in transformed plants are still time-consuming, and can be quite expensive. The objective of this study was to search for a simpler method to screen for transgenic plants. The infiltration of kanamycin (100 mg/mL) into tobacco leaves resulted in conspicuous chlorotic spots on the non-transgenic plant leaves, while no spots were seen on the leaves of transformed plants. This reaction occurred regardless of age of the tested plants, and the method has proven to be simple, fast, non-destructive, relatively cheap, and reliable. These results were comparable to those obtained by the polymerase chain reaction (PCR) amplification of the transgene using specific primers. |

Twenty goats ranging from 4 to 7 months of age were divided into two groups. Group l consisted of five goats inoculated intradermaIly with 4.0X 106 virulent Corynebacterium pseudotuberculosis. Group 2 consisted of fifteen goats vaccinated intradermaIly with a single dose of 20 X 103 C. pseudo- tuberculosis. Twelve weeks later the goats in group 2 were reinoculated with 4.0 X 106 virulent organisms. AlI goats from group l developed abscesses at the site of inoculation, in the draining lymph nodes, as weIl as in internaI viscera and or lymph nodes. None of the go!ts in group 2 developed abscesses in their external lymph nodes or viscera but one goat had a small abscess in a single mesenteric lymph node and one goat in a retropharyngeal lymph node. Ten of the 15 goats from group 2 had smaIl abscesses at the site of inoculation which developed after the challenge inoculation. It was concluded that small doses of live C. pseudotuberculosis stimulated an immune response capable of preventing the development of abscesses in the draining lymph node as weIl as protected against the development of internai abscesses. Résumé: Vingt moutons ages de 4 a 7 mois sont divises en due groupe. Le primier groupe est constitue de cinq moutons aux quels ont a injecte une 4.0 × 106 de Corynebacterium pseudotuberculosis. Le deuxieme groupe est constitue de quinze moutons auxquels on a injecte une dose unique de 20 × 103 de C. pseudotuberculosis. Apres douce semaine 1 experience tot renouvelee avec une dose 4.0 × 106 du meme produit. Tous les moutons du groupe 1 developperent un abces au point d inoculation, dans les ganglions lymphatiques de drainage aussibien que dans les visceres et (ou bien) les lymphatiques. Avcun des mouton de groupe deux ne developpa d abces dans les ganglions lymphatiques external ou dans les viseres, mais un moutons developpa un un petit abces dans un mesenteric ganglion et autre le developpa dans le retropharyn ganglion lymphatique. Dix des 15 moutons du groupe 2 developperent un abces au point d inoculation qui produisit une reaction. En conclusion les petites doses de C. pseudotuberculosis vivantes stimulees en reponse immunitaire capables de prevenir le developpement d'abces dans le drainage de la ganglion lymphatique aussi bien que de proteger contre le developpment d'abces interieur.
