The Eucalyptus genus comprises over 500 species, among which are timber species of rapid growth, commonly grown in the world for use in pulp and paper industry. Aiming a sustainable use of timber, supplying the demand and meeting the requirements of industries, the breeding programs seeks, among other interest, to improve the wood quality, lower lignin and extractive and increase the density of wood. Therefore, it is interesting to identify genetic markers that characterize the major events of xylem differentiation once it is probably a key factor to determine some properties of wood and therefore influence their performance and industrial value. Aiming to identify genes specifically involved in the synthesis and characterization of the wood, three suppressive subtractive libraries (Suppression Subtractive Hybridization - SSH) were constructed. The libraries were built using four xylem samples with contrasting characteristics to wood quality, extracted from trees of a eucalyptus population from commercial company Suzano. A total of 686 DNA sequences were obtained, being 328 contigs in BSEUC1, 397 sequences and 79 contigs in BSEUC2, 449 sequences and 90 contigs in BSEUC3. Among the most unigenes representative, annotated by the program Blast2GO versão1.2.7 in BSEUC1, was identified a transcription factor, which accounted for 44 repetitions in the library, and a protein of unknown function, which corresponded to 28 repetitions. Were also identified genes, 30.2% of the total, which were not repeated in the library and many with unknown function. In BSEUC2 a hypothetical protein was identified, which corresponded to 58 repeats, a protein associated with senescence and with 54 repetitions of an ADP ribosylation factor with 51 repetitions. In BSEUC3 was identified in a heat shock protein, corresponding to 59 repetitions, and a transposase with 54 replicates. Sequences with single repetition also appeared in the latter two libraries, like sequences with unknown function. For expression analysis by qPCR and validation of the most representative sequences from each library were selected 9 genes of BSEUC1, 5 genes of BSEUC2 and 8 genes of BSEUC3. The conservation and pre-breeding of natural populations of A. coriacea depend on genetic divergence studies and require knowledge about its characterization and the possible identification of superior genotypes. |
