Abstract: Studies on the regulation and modulation of starch composition and structure in store organs have been traditionally done based on mut ations in a specific step on the starch biosyntheses pathaway. Several kinds of mutants on starch biosyntheses have been described in organs surch as seeds and tubers, but nome have been found in store root like the one in cassava. We are reporting the discovering of storage root starchless clone in cassava identified in wild and cultivated by rural community in Amazon. Iodine solution was used to test for the presence of starch in storage root of several Manihot species, including M. esculenta subsp esculenta. Among several clones of cassava tested in the region, only the clone CAS036 showed low starch presence. CAS036 was also found to be cultivated in rural community in agricultural systems like Indian cultivation, "roças cultivation", and garden cultivation, all for specific needs as sugar source for dessert. Several plants with similar characteristics were collected for biochemical and molecular characterization. Our results have indicated that the CAS036 clones store over 100x more sugar in the form of glucose in the bulk of the store root than other commercial clones. It was also observed that polysaccharides in CAS036 is less than 2% (dry matter base). Biochemical and molecular characterization are being carried out and will be discussed based on the possible utilization of this clone for sugar inter-conversion, starch structure modification and physiological consequences of store this amount of osmotically active sugar.
