Populações de açaizeiros na Amazônia são abundantes, possuem importância econômica e têm sido foco de diferentes estudos. Os açaizeiros destacam-se por produzir um importante alimento: a ?polpa de açaí?? extraída dos frutos. Nos últimos anos, a demanda pela polpa aumentou de forma expressiva por possuir características energéticas, nutritivas e altos teores de fibras e antioxidantes. A extração dos frutos é baseada no extrativismo e por isso, práticas de manejo têm sido recomendadas para uma coleta sustentável. Para se recomendar taxas sustentáveis de coleta dos frutos, é fundamental o levantamento de informações sobre a variabilidade genética das populações naturais, além do monitoramento sobre o recrutamento e a dinâmica populacional. A transferibilidade dos microssatélites tem trazido bons resultados entre locos desenvolvidos para Euterpe edulis e aplicados para estudos em Euterpe oleracea e Euterpe precatoria. Foi caracterizada ampla diversidade genética entre os acessos do banco ativo de germoplasma de E. oleracea situado na Embrapa Amazônia Oriental. Já nos Estados do Amazonas e do Acre, os valores encontrados foram de diversidade genética intermediários a baixos em populações de E. precatoria, com significativa estrutura genética espacial. Dados moleculares foram usados para identificação de uma coleção nuclear e para direcionar estratégias de conservação e coleta de sementes para conservação in e ex situ em E. precatoria. A genética molecular é uma ferramenta valiosa e está disponível para diversas aplicações em açaizeiro. A estimativa de parâmetros genéticos tem contribuído a cada dia para o conhecimento da genética da cultura. Populations of açaí palm in the Amazon are abundant, have economic importance and have been the focus of different studies. The açaí palm stands out for producing an important food: the "açaí pulp" extracted from the fruits. In recent years, demand for pulp has increased significantly because it has energy, nutritional characteristics and high levels of fiber and antioxidants. The extraction of the fruits is based on the extractivism and therefore, management practices have been recommended for a sustainable collect. In order to recommend sustainable rates of fruit collect, it is essential to collect information on the genetic variability of natural populations, as well as monitoring on recruitment and population dynamics. The transferability of the microsatellites has brought good results among loci developed for Euterpe edulis and applied for studies in Euterpe oleracea and Euterpe precatoria. It was characterized a wide genetic diversity between the accesses of the active germplasm bank of E. oleracea located in Embrapa. In the states of Amazonas and Acre, the values found were of intermediate to low genetic diversity in populations of E. precatoria, with a significant spatial genetic structure. Molecular data were used to identify a core collection and to direct conservation and seed collect strategies for in situ and ex situ conservation in E. precatoria. Molecular genetics is a valuable tool and is available for several applications in açaí palm. The estimation of genetic parameters has contributed every day to the knowledge of the genetics of the culture. The cellular uptake of dsRNA after dietary exposure is critical for RNAi efficiency; however, the mechanism of its uptake in many insects remains to be understood. In this study, we evaluated the roles of the endocytic pathway genes Clathrin heavy chain (Chc), Clathrin adaptor protein AP50, ADP ribosylation factor-like 1 (Arf72A), Vacuolar H+ ATPase 16 kDa subunit (Vha16), and small GTPase Rab7 and putative sid-1-like genes (silA and silC) in RNAi response in western corn rootworm (WCR) using a two-stage dsRNA exposure bioassay. Silencing of Chc, Vha16, and AP50 led to a significant decrease in the effects of laccase2 dsRNA reporter, indicating that these genes are involved in RNAi response. However, the knockdown of either Arf72A or Rab7 did not suppress the response to laccase2 dsRNA. The silencing of the silC gene did not lead to a significant reduction in mortality or increase in the expression of V-ATPase A reporter. While the silencing of the silA gene significantly decreased insect mortality, significant changes in V-ATPase A expression were not detected. These results suggest that clathrin-dependent endocytosis is a biological mechanism that plays an important role during RNAi response in WCR adults. The fact that no definitive support for the roles of silA or silC in RNAi response was obtained support the idea that RNAi response varies greatly in different insect species, demanding additional studies focused on elucidating their involvement in this mechanism. |
