The present research seeks to elucidate the feasibility of chitosan (CHT) in the induction of water deficit tolerance in different maize hybrids, contrasting tolerance to water restriction, tolerance and sensitivity. The maize plants were subjected to water deficit and foliar application of different chitosan doses (60, 100, 140, and 180 mg L-1) at the pre-flowering growth stage and evaluated during the stress period of fifteen days. To understand the induction behaviour of the tolerance to water restriction, biophysical parameters, such as water potential, relative water content and chlorophyll content, gas exchange, and biochemical assays, were quantified based on the activity of SOD, CAT, APX, and PAL antioxidant enzymes, lipid peroxidation activity and hydrogen peroxide content. Among the treatments, maize plants subjected to chitosan foliar application at a dose of 140 mg L-1 presented similar behavioural responses to plants under favourable irrigation conditions. Such positive responses are related to the high degree of activity of antioxidant enzymes, gas exchange and low levels of lipid peroxidation and hydrogen peroxide. The results support the potential use of CHT to increase tolerance to water stress. |

Resumo: Há uma necessidade de adotar estratégias de conservação para as populações naturalizadas de animais domésticos para assegurar a manutenção destas para o futuro. Este estudo visa contribuir para o conhecimento da variabilidade genética por meio de marcadores de microssatélites em caprinos naturalizados Moxotó, Canindé e Marota. Foram amostrados 124 animais adultos dos grupos Moxotó, Canindé e Marota dos núcleos de conservação da Embrapa. O DNA extraído foi amplificado mediante a reação em cadeia polimerase (PCR) e genotipado através do programa Fragment Profile (Amersham Bioscience). O método UPGMA (Unweighted Pair Group Method With Arithmetic Mean) foi utilizado para construção do dendograma com bootstrap (1000 repetições) pelo programa TFPGA (Miller, 1997). O dendograma agrupou no primeiro nódulo as populações Moxotó e Canindé e o bootstrap apresentou 75% de similaridade neste nódulo. No segundo nódulo houve a inclusão das três populações e obteve bootstrap de 100%. O número de loci suportando cada nódulo foram quatro e cinco, respectivamente. O método UPGMA, utilizando as distâncias de Nei (1978), permitiu o discernimento das populações caprinas com base em dados moleculares.
[Genetic distance in naturalized goats using DNA microsatellites].
Abstract: There is a need to adopt conservation strategies for naturalized populations of domestic animals to ensure their maintenance for the future. The objective of this study was to determine the genetic variability in naturalized Brazilian goat populations using microsatellite markers. Samples of 124 Moxotó, Canindé and Marota adult animals were obtained from conservation flocks maintained by Embrapa. The extracted DNA was amplified by chain polimerase reaction (PCR) and genotyped using the Fragment Profile program (Amersham Bioscience). The Unweighted Pair Group Method With Arithmetic Mean method (UPGMA) was used for construction of the dendogram with bootstrap (1000 repetitions) of the TFPGA program (Miller, 1997). The Moxotó and Canindé groups were included in the first nodule of the dendogram with a bootstrap value of 75%. All three groups were included in the second nodule with a bootstrap value of 100%. The loci number supporting each nodule was four and five, respectively. The method UPGMA with molecular data, using Nei's distances (1978), allowed for the discernment of these goat populations.
